No dont restart the OS, just stop the Services, I dont know if you are running NT or 2000 but there should be a "Services" applet in control panel or in "Administrative Tools" In Services you need to stop IIS Admin Service, this will inturn stop WWW, SMTP and FTP. replace the DLL then Start all the services back. No restart of the OS, Forget about using the inuse.exe, it just sets files to be replaced on the next reboot.
